Piris has signed a two year contract with the Brazilian side, and Cerro Porteno will receive a fee in the region of €3 million for the 22-year-old.
Speaking to the club's official website, Sao Paulo vice-president John Paul de Jesus Lopes said: "For months we have been working quietly on this player and to see the deal completed makes us very happy.
"He is a young athlete with a lot of potential. He will definitely add great quality to our group," he added.
The defender is currently with the Paraguay squad ahead of the Copa America final against Uruguay on Sunday. He has played twice in the competition so far, in the group stage draw against Ecuador and the semi-final triumph against Venezuela.
Piris was also involved in both legs of Cerro Porteno's Copa Libertadores semi-final defeat against Santos in May.
